First introduced by Dragon, the Holy Knights have only grown in power and importance since, and now, they're playing an active role in the Elbaf arc of the story. The Holy Knights have already been seen fighting, and it is quite clear that they're monsters in combat, but their biggest strength is that they are immortal and can regenerate infinitely.

The Holy Knights have distinct abilities in One Piece. When they were first shown in Elbaf, it became clear that they hold tremendous power. Gunko was the first one to show her powers, and it was clear that she held the ability of arrows. Later, it was confirmed that Gunko has the Aro Aro no Mi. Similarly, Shamrock's sword was revealed to have the power of the Cerberus. Later, two more Holy Knights came, and one of them, Sommers, was shown to hold the power of the Iba Iba no Mi, while the other, Killingham had the ability of the Ryu Ryu no Mi, Model: Kirin.

The Devil Fruit powers of the Holy Knights are immense, and fans also know very well that they use two types of Haki, at the very least, with some using all three. However, their biggest strength isn't their Devil Fruit power. They're immortal and they can regenerate from all damage that is inflicted upon them. This makes them invincible. However, as confirmed by Gaban, they can indeed be defeated.

In One Piece chapter 1152, Scopper Gaban, the left arm of the Pirate King, confirmed to the fans that the biggest weakness of the Holy Knights and the Five Elders, and even Imu, is Conqueror's Haki. This means that using this power can bypass their immortality and ensure they take full damage. Fans also saw Gaban injuring Sommers to the point where he couldn't regenerate until Imu came along.

That said, Gaban also confirmed that just using Haki isn't enough. It is how you use Conqueror's Haki that matters. This means just coating techniques in Conqueror's Haki isn't enough at all and that one needs to specifically use it to target certain elements, perhaps something like the soul.

Who Can Counter The Holy Knights?

At this point in time, only Gaban, Luffy, Zoro, and Loki can counter the immortality of the Holy Knights and beat them. However, Sanji appears to be incredibly close to gaining this power as well, as was hinted at by Oda in One Piece chapter 1152, and long before that as well. It is only a matter of time before he awakens this ability.
